Evaluation of Viral RNA Recovery Methods in Vectors by Metagenomic Sequencing

Joyce Odeke Akello, Stephen L. Leib, Olivier Engler and Christian Beuret

Identification and characterization of viral genomes in vectors including ticks and mosquitoes has challenges. One such challenge is the ability to efficiently recover viral RNA which is typically dependent on sample processing. Six different extraction methods were evaluated for recovering viral RNA in vectors and the ROVIV-method (Recovery of Viruses in Vectors (Method 5)) - that uses  QIAzol, Proteinase K and silica magnetic beads – performed best in viral RNA recovery and identification in vectors.
